Program to generate a protocol for DMS probing from a defined template.
- Requires Python 3
- Requires json and tkinter libraries (included in most Python installations)
python build_protocol.py
- choose/enter values for your experiment
- choose output location
- hit "generate" button
Hardcoded values can be edited in config.json. New plate types can be added to the dropdown menu by editing config.json under the "plate_type" field. The first entry contains the name of the plate, the second is the volume of 2% DMS you intend to probe it with.
It is recommended to enter a small excess of DMS and a large excess of DTT. The excess DTT is used for quenching hardware, larger volume experiments require relatively less excess to quench hardware.
The template can be edited, the following values are replaced with calculated numbers: VOL_DMS: volume of DMS (microliters) added to create a 2% DMS solution MASS_DTT: mass of DTT (grams) required to create a solution that is 5X molar excess to an equivalent volume of 2% DMS VOL_ETOH: volume EtOH (milliliters) required to create a 25% EtOH/75%DPBS buffer for the 2% DMS VOL_DPBS_DMS: volume DPBS (milliliters) required to create a 25% EtOH/75%DPBS buffer for the 2% DMS VOL_DPBS_DTT: volume DPBS (milliliters) required to create the buffer for the DTT solution VOL_PLATE_TYPE: volume of 2% DMS (milliliters) for probing the given plate type NUM_SAMPLES: number of samples the experiment is being run on
